Sustainable Alternatives To Paper Towels
Prepare a moist chamber using a large rectangular plastic box with lid (e.g., a refrigerator storage box). Place several sheets of hand paper towels at the bottom of the box and wet with distilled water. On top of paper towels, place some plastic rods (diameter 0.8-10 mm) to support a container for the membrane. Tightly close the box with lid and place into an incubator at 37 C.
At this point, the mouse may be disinfected. The disinfectant also serves to wash off any loose hairs and mats down hair on an unshaven mouse for ease of examination. After letting some of the disinfectant drain from the mouse back into the container, place the mouse on one to two layers of absorbent paper towel on a cork board. The cork board should be approximately 14 cm x 21.5 cm in size, which is large enough for two mice, yet easy to move about during the necropsy. It should be at least 1.0 cm thick. The intestines should be collected first because autolysis begins quickly in these organs. Grasp the cecum with forceps, lift it up and cut away the small intestine at the rotund sac, where the ileum meets the cecum. This is the smaller of the two intestines joining at the cecum. Drop the cecum and grasp the severed end of the ileum, pulling it gently as the small intestine begins to unravel. Carefully cut any mesenteric tissue that provides resistance. Be sure not to puncture or...
The donor mouse is euthanized by carbon dioxide or cervical dislocation. The carcass is soaked with disinfectant (70 alcohol or quaternary ammonium) and placed on a paper towel. In the rare event that the donor mouse is not to be euthanized, procedures for survival surgery and ovariectomy are followed. Ovaries are excised from the euthanized donor mouse, using sterile instruments and aseptic technique. Either of two alternative approaches may be used to excise ovaries from the donor.
Ordinary glass cleaner used sparingly and wiped away with paper towels is used before each series of photographs and frequently during the shoot to reduce dust that will settle on the glass. Once the subject is arranged on the glass, a small wad of moistened paper towel can be used around the edges of the specimen for last minute touch-ups. Cans of compressed air or inert gases, used to blow dust out of the field of view, are useful but not necessary. If used, point the tip away from the
